YES:
Peerblock will block some IP addresses (the categories that you choose to block)
I believe you are asking will it keep me safe from MPAA/RIAA ect pinging/tracking my downloads/uploads. This is where we get to the no part of the answer
NO:
Peerblock is simply just a giant library of known IP address. The key word here is KNOWN. If the MPAA/RIAA ect wanted to track you they would just use an IP address that is not in this giant library/database (peerblocker). And yes they know which of their IP address are blocked and which are not (they have access to all peerblockers as well)

My Opinion:
Peerblocker is just a waste of RAM. It can cause lots of firewall problems and can lead to further security issues. If you are trying to be safe about downloading the best thing to do is to find a download site that is secure. What I mean by this is members ONLY and to become a member an invite is required. This helps keep torrents/servers secure from others tracking uploads/downloads. (A majority of people that are caught illegally uploading are done so because the MPAA/RIAA just download the file themselves and then tracks and logs the IP addresses. They have software to do all this for them. You can even do this yourself using the trackers and peers section of each file you have in uTorrent)
